NAD+: specifications, literature and price per milligram

Not a peptide, and sold in vial masses fifty to a hundred times larger than one. Per-milligram pricing is not a nicety here, it is the only way to read the market.

What NAD+ is

Nicotinamide adenine dinucleotide is a coenzyme found in all living cells, where it acts as an electron carrier in redox reactions and as a substrate for enzymes including sirtuins and poly(ADP-ribose) polymerases. It carries CAS 53-84-9 and a molecular weight around 663.4 g/mol.

It is a nucleotide cofactor, not a peptide, and it is cataloged alongside peptides in this market for commercial reasons rather than chemical ones.

What the published literature examined

Published literature, largely from rodent models, has described declines in tissue NAD+ concentrations with age and has examined precursors such as nicotinamide riboside and nicotinamide mononucleotide as experimental means of raising NAD+ (Rajman et al., Cell Metabolism 2018).

In humans, a randomised placebo-controlled crossover study examined the tolerability of oral nicotinamide riboside and measured blood NAD+ concentrations in healthy middle-aged and older adults (Martens et al., Nature Communications 2018). Note that this examined a precursor taken orally, which is a different subject from the material on this page.

Price per milligram

The 1000mg vial is $95.99 list, or $0.10/mg. The 500mg vial is $57.99 list, or $0.12/mg. Both figures sit beside the vial price on the product page.

Large-mass vials are where per-milligram pricing does the most work, because a difference in vial price is a rounding error at one scale and a doubling at another.

NAD+ belongs to a redox pair

The plus sign is part of the chemistry. NAD+ is the oxidized member of the NAD+/NADH redox pair. Enzymatic reactions can transfer reducing equivalents between the two forms, so a paper may measure NAD+, NADH, their ratio, or a wider metabolite panel. Those are related measurements, not interchangeable labels.

NAD+ also serves as a consumed substrate for enzyme families including sirtuins and poly(ADP-ribose) polymerases. That biochemical role is different from its role as an electron carrier. A careful overview states which role an experiment is investigating rather than placing every NAD-associated pathway under one outcome claim.

Because NAD+ is a nucleotide cofactor rather than a peptide, peptide-specific shorthand can mislead. Sequence purity and peptide mapping are not the relevant terms. The specification should name the correct chemical form, expected mass, quantitative method and related nucleotide impurities.

Direct NAD+ is not the same subject as an NAD+ precursor

Much of the human literature commonly linked from NAD+ pages studies nicotinamide riboside or another precursor. A precursor is converted through metabolic pathways; it is not the same material as an NAD+ vial. A finding about oral precursor exposure cannot be presented as a finding about direct NAD+ research material.

The 2018 randomized crossover study by Martens and colleagues measured the tolerability of oral nicotinamide riboside and changes in blood NAD+-related measures in healthy middle-aged and older adults. The design is useful and the human data are real, but the intervention identity must remain attached to the result: nicotinamide riboside, not NAD+.

This distinction prevents a common citation error. A page can discuss the wider NAD metabolism field while still stating that the cited clinical study did not examine the catalog material on the page.

What a biological NAD measurement means

A reported NAD concentration depends on sample type, collection, extraction, assay and timing. Whole blood, plasma, a cell fraction and a tissue sample are different matrices. A change in one matrix should not be described as though it measured every compartment.

Pre-analytical handling can matter because redox metabolites are chemically active. A paper's method section should explain how quickly a sample was stabilized, what internal standards were used and which metabolites the assay separated. Without those details, a clean percentage change may hide a noisy measurement process.

A strong summary therefore names the matrix and analyte. 'Blood NAD+ under this assay' is more informative than 'NAD levels,' because it tells a later reader what was actually measured.

Reading precursor trials without borrowing their conclusion

A controlled precursor trial can answer whether the tested precursor changed its defined laboratory measures in its enrolled population over its stated period. It cannot by itself answer whether another precursor, another chemical form, another route or a different population would produce the same measurement.

The comparator and endpoint hierarchy also matter. Safety observations, a biochemical endpoint and an exploratory functional endpoint should not be presented as though they were all primary findings. Registration, prespecified analysis and complete reporting help a reader see which questions the study was designed to answer.

This page uses precursor work to explain why NAD metabolism is studied. It does not use that work to make a claim about the material sold here.

Analytical questions for an NAD+ vial

A useful release record starts with identity, purity and quantity, but the methods are not automatically the same as those used for a peptide. The report should name the reference standard and distinguish NAD+ from related nucleotides and degradation products under the chosen chromatographic method.

  • Identity: an instrumental comparison tied to the intended chemical form.
  • Purity: a chromatographic result with trace, method and integrated peaks.
  • Measured mass: the quantitative amount reported separately from purity.
  • Nominal mass: the label claim used for the purchasing comparison.
  • Lot linkage: a sample trail connecting the test result to the inventory record.

A large measured mass does not reduce the need for identity. A thousand milligrams of the wrong or degraded material would still be the wrong result.

Why purity and measured mass need separate columns

The 1000mg scale makes the distinction visible. Purity is a composition result under an analytical method. Measured mass is a quantity result. A vial can be close to its nominal mass while showing a lower composition result, or show a high composition result while the quantity differs from the label.

For price comparison, start with the nominal label because that is what the offer promises. For release and investigation, retain the measured result and its uncertainty. Do not silently replace the label with the observed number or convert one historical sample into a permanent claim about the product.

A restrained way to publish historical testing

Historical third-party results can show how a sourcing program has been evaluated, but they do not certify a future Effect lot. The public table therefore labels them as sample results and exposes only product, labeled mass, measured mass and reported purity. It does not imply that the tested sample is the inventory a buyer will receive.

The complete source reports remain private because they contain verification and report details the owner has chosen not to redistribute. Privacy of the source record does not change the scope of the public claim: it is a limited summary, not a downloadable lot-matched certificate.

Map the NAD metabolome before choosing an endpoint

NAD biology includes oxidized and reduced dinucleotides, phosphorylated counterparts and precursor or breakdown metabolites. An assay that reports one member of that network may be excellent for its target while saying nothing direct about the others. The analyte list should be explicit.

Ratios can answer a different question from absolute concentrations. A stable total with a shifted ratio, or a changed concentration in one compartment, should not be summarized as a universal change in 'NAD.' The matrix, analyte and calculation all belong beside the result.

Targeted mass-spectrometry panels, enzymatic cycling assays and other approaches have different separation, specificity and calibration properties. Comparing studies requires reading the analytical method as well as the biological design.

Pre-analytical handling can become part of the result

Metabolites can change during collection and processing. Time to stabilization, temperature, extraction procedure, freeze-thaw history and internal standards can influence the reported concentration. These are not housekeeping details when the measured system is chemically dynamic.

A paper that reports a precise concentration should therefore be read with its sample workflow. If two studies used different matrices or handling protocols, their numbers may not share a common scale even when both are labeled NAD+.

For reproducibility, record the workflow rather than copying only the final concentration. A later group needs the collection and extraction decisions to recreate the measurement.

A precursor evidence map

Precursor studies can be organized by chemical identity, route, population, duration and measured matrix. Nicotinamide riboside, nicotinamide mononucleotide and niacin-related materials enter the wider network by different paths and should not be collapsed into one intervention category.

The first endpoint may be exposure or a change in a blood metabolite. Later exploratory endpoints answer separate questions and often have less statistical power. A summary should preserve which endpoints were primary, which were secondary and which were exploratory.

This map is useful background for NAD metabolism, but it does not establish identity or performance for direct NAD+ research material. That requires its own analytical and experimental record.

Stability claims require their own data

A supplier may give storage or handling statements, but those statements should come from stability data for the defined material and container rather than from the general reputation of the molecule. Related nucleotide products may form under conditions that a simple appearance check will not resolve.

A stability program defines time points, conditions, analytical methods and acceptance criteria. A single release result is a snapshot and cannot establish a shelf-life claim by itself.

Effect's public historical table is not a stability program. It reports limited observations from supplied sample records and does not extend those observations across storage time or future lots.

Chemical identity in a category built around peptides

NAD+ appears in a peptide catalog for commercial convenience, but its specification should follow nucleotide chemistry. Peptide-sequence language, amino-acid analysis and peptide purity shorthand can make a page sound technical while failing to define the actual analyte.

The identity file should name the oxidized dinucleotide form, formula or defined salt basis, expected mass and reference material. The chromatographic method should distinguish relevant related nucleotides or degradation products to the extent required by the specification.

This category distinction also matters when comparing vendors. A listing for NADH, a precursor, a mixture or an unspecified 'NAD' product is not a matched price row for NAD+. Normalize the identity before normalizing the price.

A transparent conclusion can remain limited

A long research page does not need a dramatic verdict. The defensible conclusion is that NAD metabolism is a substantial field, much of the accessible human intervention literature concerns precursors, and direct NAD+ research material still requires its own chemical and analytical record.

That conclusion leaves several questions open by design. It does not infer a biological result from vial purity, does not infer a vial's identity from a precursor study and does not extend a historical sample result to future lots.

Those separations make the page easier to update. New biological studies can be added to the evidence map, while new inventory results can be added to the lot record without confusing the two.

Frequently asked

Is NAD+ a peptide?
No. It is a dinucleotide coenzyme. It is cataloged alongside research peptides for commercial reasons rather than chemical ones.
Why compare NAD+ per milligram rather than per vial?
Because vial masses in this category run from 500mg to 1000mg and higher. Vial price alone tells you nothing about which is cheaper.
